He took money from a till before escaping on foot.
Nobody was injured but a female customer was treated by ambulance staff for breathing problems.
The man wanted by police is white, 6ft tall and of stocky build.
He was wearing a black beanie hat with a white stripe on the bottom, a black scarf around his face, dark jeans and a black zipped topped. Enquiries are ongoing to trace him.
Detective Inspector Allison Woods from Wirral CID said: "This would have been a shocking incident to witness.
"Staff and customers came face to face with a man brandishing a knife in broad daylight, with no regard for the fear that this causes.
"We are determined to find the person responsible and would urge anyone who saw the incident or the man making off from the bank just before 9.30am to contact police.
"We are also particularly keen to speak with any customers in the bank at or around this time, or anyone in the community who recognises his description."
Anyone with information or who recognises the man should call Wirral CID on 0151 777 2263 or Crimestoppers anonymously on 0800 555 111.
